This homepage presents information from the Research Laboratory at the Department of Haematology, Aalborg Hospital, Aarhus University.

The Research Laboratory was established during 2005-2007. Competent staff members have been employed in accordance with the research strategy within a visible organization and management structure.

The scientific goals are to achieve new insight on the mechanisms concerning the haematological cancer diseases with focus on the human B-cell malignancies and associated disorders.

The laboratory is a section of the Haematology Research Department and is located at Aalborg Hospital Science and Innovation Centre with core functions for gene array and protein array technologies as well as core function for multiparametric flow cytometry and cell sorting. The Bio-Laboratory (Biobank), the Clinical Trial Unit and the Clinical Department are located at Medicinerhuset (the Medical Centre).

Annual Meeting in DAHFO

On 16-18 March Dansk Hæmatologisk Forskerskole, DAHFO, held an annual meeting at Hindsgavl Slot with Karen Dybkær Sørensen and Hans E Johnsen as organizers.

The title of the meeting was “State of the Art – Pathogenesis, Novel Technologies and Clinical Trials in Haematological Malignancies”.

15 invited professors and associate professors, with great knowledge within the areas pathogenesis for leukaemia, lymphoma and bone marrow cancer and methods for a.o. diagnostics and general research as well as clinical protocols, gave lectures.

The programme is available at the website of Dansk Hæmatologisk Forskerskoles webside, www.dahfo.dk.

19 research students, PhD students and postdocs participated in the meeting, among them Liesbeth Bieghs, Maria Domingo, Steffen Falgreen Larsen, Kim Bergkvist, Thomas Urup, Malene Kjeldsen, Julie Støver Bødker and Maria Bro Kloster from our department. Martin Bøgsted, Mette Nyegaard, Alex Schmitz, Karen Dybkær Sørensen and Hans Erik Johnsen from our department gave lectures.

The annual meeting of Dansk Hæmatologisk Forskerskole will be held again in 2012 same time and place in connection with the annual meeting of Dansk Hæmatologisk Selskab.

The NMSG Workshop on "Translational Research in Multiple Myeloma was held at Hotel Hilton Copenhagen Airport the 28th October 2010.

PRIMA study

The Department of Haematology, Aalborg Hospital, has included patients into the PRIMA study just published in the respected journal Lancet Oncology.
The study demonstrated improved progression-free survival in follicular lymphoma patients after Rituximab maintenance treatment.

Scholarship from "Kræftens Bekæmpelse"

Master student in Medical Biotechnology, Annie Bendtsen, has received a scholarship of 100.000 DKK from "Kræftens Bekæmpelse" for the project: "Analysis of Pax5’s promoters and alternative transcripts in diffuse large B-cell lymphoma".

The main objective is to clarify the role of Pax5’s two promoters and resulting alternative transcripts in normal B-cells and in ABC and GCB DLBCL cells.
The aim is to determine the level of expression of the two transcripts of Pax5 by q-PCR and gene expression microarray. To determine the level of the two protein isoforms of Pax5 by Western blot. To determine the methylation level of the promoter sites by methylation sensitive high resolution melting (MS-HRM). To investigate the alternative splicing of Pax5 by exon microarray.

More than 1500 patients included in the Haematology Biobank

As of November the 15th, the Haematology Biobank at Aalborg Hospital has registered more then 1500 patients, with close to 12.000 individual samples. The archived patient material includes blood, plasma, serum and bone marrow samples as well as biopsies.

Two New PhD Students

Steffen Falgreen Larsen and Liesbeth Bieghs have been granted a PhD mobility stipend and are enrolled as PhD students at the Department of Haematology by December 1st 2010.

Steffens PhD project is entitled "Statistical Models Studying Potential Biomarkers for Prediction of Chemotherapy Outcome" and the main supervisor is associate professor Martin Bøgsted, Department of Haematology, Aalborg Hospital, Aarhus University Hospital.

Liesbeths project is entitled "The IGF system in multiple myeloma. Exploring diagnostic and therapeutic potentials" and the main supervisor is associate professor Mette Nyegaard, Department of Haematology, Aalborg Hospital, Aarhus University Hospital.

European Myeloma Network Flow Cytometry Worskshop, 2010

On the behalf of the European Myeloma Network (EMN), our group together with the group from Alberto Orfao (Salamanca, Spain), have organized a workshop focused on minimal residual disease monitoring in the European Intergroup Trial EMNIT. The workshop was held in Salamanca, Spain on the 23rd of October and ca. 20 scientists from 10 different European countries have participated including Hans E. Johnsen and Alexander Schmitz from our group. The workshop was supported from Cytognos (www.cytognos.com).

On 24 September 2010 Alexander Schmitz made an oral presentation of  "FACS-SORTED B-CELL SUBPOPULATIONS FROM NORMAL HUMAN SECONDARY LYMPHOID TISSUE; FIVE SUBSETS WITH CONVERGENT GENE EXPRESSION PROFILES AND PHENOTYPE" at the 10th Euroconference on Clinical Cell Analysis in Valencia/Spain, after having been selected for oral presentation from the accepted poster abstracts.
Participation was supported by the Danish Cancer Society (Kræftens Bekæmpelse).

Conference on Strategic Research

Conference on strategic research within cancer treatment Professor Hans E Johnsen from the Department of Haematology, Aalborg Hospital, Aarhus University Hospital, was among the speakers at a conference on strategic research within cancer treatment, bearing the title “Cancer – from treatment of diagnoses to treatment of citizens”
The conference took place on 2 September 2010 at the county hall at Christiansborg and was arranged by the Strategic Research Council (Det Strategiske Forskningsråds programkomiteer for Sundhed, Fødevarer og Velfærd samt for Individ, Sygdom og Samfund)

At the conference leading cancer scientists were telling how they - by means of grants from the Strategic Research Council - research into the individual patient’s specific needs for treatment.

Participation in the AMEE conference 2010

Simon Mylius has participated in the AMEE conference (Association for Medical Education in Europe) from 4 to 8 September.
AMEE is a worldwide organization with members in 90 countries. The conference was held in Glasgow, Scotland, and lasted 5 days with courses, workshops, symposiums, poster sessions and more, discussing the actual and future perspectives of medical education in global and local settings. Simon participated as a representative from the Medical Students Association at Aarhus University.
Participation was supported by the Faculty of Health Sciences at Aarhus University.

New Employee at the Haematology Biobank

The internship that was granted to Stig Castberg has been extended to public wage subsidy, which is a part-time position at the Haematology Biobank.

Stig graduated in 2009 from the Institute of Physics and Nanotechnology at Aalborg University. His master was on the subject 'Expression of an Indolicidin Analogue in the Yeast Pichia Pastoris'. During his education he has obtained laboratory experience with bacteria and yeast cells as well as different peptides and proteins.

Stig will be preparing the daily samples of the Biobank.

Hydroxyurea has been used for treatment of blood cancers since the early nineteen-sixties. We have now identified the hydrogen peroxide decomposing enzyme catalase as a novel direct and biologically significant target of hydroxyurea.

More than 1200 patients included in the Haematology Biobank

As of April 26th, more than 1200 patients are registered in the Haematology Biobank at Aalborg Hospital. The archived patient material includes blood, plasma, serum and bone marrow samples as well as biopsies. In total, the Haematology Biobank holds close to 10.000 individual sample vials.

New Intern at the Haematology Biobank

On Monday 12 April Stig Castberg, the newly graduated bio-engineer,  started a four week internship at the Haematology Biobank.

Stig graduated in 2009 from Institute of Physics and Nanotechnology at Aalborg University. His master was on the subject 'Expression of an Indolicidin Analogue in the Yeast Pichia Pastoris'. During his education he has worked in the laboratory with both bacteria and yeast cells and different peptides and proteins.

During his internship, Stig will be introduced to the daily routine of the Biobank by Biobank Manager, Linda Pilgaard.

Seminar on cancer stem cells and biobanking

On Friday March 19th Post Doc, Kirsten Fogd gave a seminar on strategies in the identification and characterization of cancer stem cells in haematology. This was followed by Post Doc, Linda Pilgaard with a seminar on biobanking and a tour of the Haematology Biobank at Aalborg Hospital.  Approximately 20 students from Industrial Medicine at Aalborg University participated in the event.

The National Medical Student Research Congress 5–7 March 2010

Simon Mylius participated in the National Medical Student Research Congress which was held at Sandbjerg Gods in the South of Jutland, Denmark, from 5-7 March 2010.

At the Congress more than 80 medical students presented their scientific work. There were 7 oral sessions and many posters, all in English.

The research projects consisted among others of epidemiological, neurological, cardiological and immunological projects.

As Simon Mylius had arranged the congress together with the other board members of the Society of Medical Research Students he participated in the congress.
The congress was a success.

Our collaborator Cand Scient PhD Stig U. Andersen from the Department of Molecular Biology, University of Aarhus presented data on "A novel direct target of hydroxyurea - identification and clinical investigation"

Abstract: The Hydroxyurea has a cytoreductive effect that is beneficial in a number of hematological malignancies, but many of these eventually develop hydroxyurea resistance. To address this issue, a plant model system was used to identify genetic lesions resulting in hydroxyurea resistance and a gene product, which directly interacts with and is inhibited by hydroxyurea, was identified.  The clinical significance of this interaction is now being investigated both retrospectively and prospectively in a collaboration between the Department of Hematology, Aalborg Hospital and the Department of Molecular Biology, Aarhus University with the aim of developing new prognostic tools to guide clinical decisions in individual cases.

Highlights from 2009


Another year has passed with a lot of activities and important changes.

Most importantly, the head of our research laboratory Cand Scient PhD Karen Dybkaer obtained a position as Associate Professor at Aarhus University, Clinical Institute.
Cand Scient PhD Mette Nyegaard has been employed as a senior scientist in molecular genetics and Cand Scient PhD Alexander Schmitz has been employed as our new full time Flow Cytometry and Sorting Manager.
Medical student Johanne Marie Holst and Stud stat Steffen Falgreen Larsen were employed part time in our statistical unit, supervised by Martin Boegsted.
Kirsten Fogd and Linda Piilgaard received their PhD from Aalborg University. Emilie Kallehauge finished her master thesis.
The technicians Hanne T. Larsen, Rikke Barsdal and Helle Høholt and the scientists Alex Schmitz, Linda Pilgaard and Kirsten Fogd visited the “Banco Nacional de ADN” and the Flow Core Unit at the University of Salamanca.


Research Assistants


In 2009 the Research Laboratory was assisted by four medical research assistants to contribute to the ongoing research. All projects made use of Affymetrix gene expression data as well as B-cell differentiation.

Jakob Virenfeldt focused on haematopoietic differentiation and their involvement in lymphoma genesis.

Katrine Gade worked with characterization of B-cell markers by flow cytometry and gene expression analysis of haematopoietic cell lines.

Sofie Hallager worked with analyzing and comparing gene expression in embryonic stem cells, B-cell lines, normal B-cells and normal tissue cells.

Jonas Svenningsen worked with identification of disease specific genes in haematological cancers by in silico analysis with the corresponding cell lines.

 


Scientific Seminar

Martin Perez-Andres stayed at the Research Laboratory for 3 months (from October to December) as a guest researcher. Martin holds a PhD in Haematology from the University of Salamanca (Spain) focused on Multiple Myeloma interaction with the immunological bone marrow microenvironment.

Martin worked in a joint-project on identification, isolation and characterization of the B cell subsets in normal individuals and patients suffering from malignant B cell diseases.

In November Martin held a presentation with the title "Immunophenotyping of haematological malignancies: Application for diagnosis and monitoring in multiple Myeloma”.

He is now back in Spain, working as Assistant Professor of Immunology at the Department of Medicine, University of Salamanca. He is interested in B-cells biology both in normal and pathological conditions.

 

 

FøSu Meeting, summer 2009

The half-yearly FøSu meeting was held in august 2009.

At the meeting the work packages 1-6 were discussed:

Work package 1:

This work package will probably result in 10-20 publications.

Work package 2:

Phase III clinical trial: A prospective randomized trial of second line therapy in lymphoma patients comparing outcome of conventional with individual DNA chip predicted experimental chemotherapy CHEPRE now includes all haematological diseases and the revised protocol has been implemented. There will be a status check when 50 patients have been implemented.

There will be a new protocol that includes Diffuse Large B-cell recidives and professor Hans Erik Johnsen is in contact with multiple centers which might be interested in international European protocol cooperation.

Steen Knudsen presented prediction on blinded retrospective Diffuse Large B-Cell samples and scheduled the next 6 months.

Work Package 3:

Malene Krag and Alex Schmitz presented FACS results from sub-population prediction and tonsil samples.

Work Package 4:

Karen Dybkaer suggested a new definition of WP4 and this was discussed.

Work Package 5 and 6:

Finn Skou Pedersen presented background, results and status. Nearly all mice have developed symptoms. A comparison between human and murine results was presented.

 

 

Summer trip to Egholm


This year’s summer trip went to Egholm and everybody enjoyed the day.

"New" staff member

Alexander Schmitz is a new staff member as of 1 January 2010. Alex holds a doctoral degree in natural science (dr.rer.nat) from Giessen University (Germany). He did a post doc at the molecular biology department (MBI), Aarhus University. Since 2002 Alex has been responsible for the establishment, management and operation of the flow cytometry unit (FCU) at MBI.

In 2007 he started working part-time at Aalborg Hospital Science and Innovation Centre (AHSIC) to establish a core function for flow cytometry and cell sorting at the Research Laboratory at the Department of Haematology, Aalborg Hospital, Aarhus University.

Alexander Schmitz will continue this work, now employed full-time as cell sorting and flow cytometry manager, providing technical and application support and expert advice, as well as holding the overall responsibility for the day-to-day running and maintenance of the flow  
analysers (BD FACSCanto2) and cell sorters (BD FACSAriaI).

Guest researcher

Martin Perez-Andres is staying at the Research Laboratory for 3 months as a guest researcher, starting from October 5th 2009. Martin holds a PhD in Haematology from University of Salamanca (Spain) focussed on Multiple Myeloma interaction with the immunological bone marrow microenvironment. New staff member

Mette Nyegaard is a new staff member as of June 1st 2009. Mette holds a PhD in Medical Genetics from Odense University, did a postdoc at Stanford Univerisity Medical School and comes from a position as Assistant Professor at the Department of Human Genetics, Århus University. Mettes special interest is genetic variation in human diseases.

Mette is employed as a senior scientist and will be focusing on identifying somatically acquired mutations and genes critical for the development of human cancers.

In house Affymetrix Cytogenetics SNP6 training

We are implementing SNP analysis at the research laboratory. In connection to this a technical expert from Affymetrix is giving a 4 days crash course for interested staff in the method involving hands-on experience with the method as well as insights into data analysis and interpretation.

Research assistants for the summer 

This summer four medical students have been employed as research assistants to contribute to the ongoing research at the Department of Haematology. Common for all projects are the use of Affymetrix gene expression data as well as B-cell differentiation:

Laboratory stay at University of Salamanca, Spain

The technicians Hanne T. Larsen, Rikke Barsdal and Helle Høholt and the scientists Alex Schmitz, Linda Pilgaard and Kirsten Fogd are visiting the “Banco Nacional de ADN” and the Flow Core Unit at the University of Salamanca. This is organized as part of our collaboration with the Cancer Research Center IBMCC at the University of Salamanca. The aim of the laboratory stay is to gain an insight into the practical and organizational aspects of the two core facilities. This will benefit the further development of the regional Biobank at the Department of Haematology, Aalborg Hospital.

Ph.D. degree received

August 29th 2008 Kirsten Fogd defended her PhD thesis “The effect of hypoxia on gene expression profiles of a human mesenchymal stem cell line undergoing osteogenic differentiation”.
The thesis was recommended for approval by a international assessment committee and November 12th 2008 the PhD title was awarded by the Academic Council at the University of Aalborg (AAU).

The thesis was written at the Laboratory for Stem Cell Research at the Department of Health Science and Technology at AAU, but Kirsten Fogd has been employed at the Department of Haematology since November 2007 as a postdoc.

The Festival of Research (Forskningensdøgn)

The Festival of Research is an annual national event organized by the Danish Agency for Science, Technology, and Innovation. It is intended to arouse public interest in and enhance public understanding of the methods, processes, and outcome of research and science. Kirsten Fogd is participating with the lecture "Når blodet går amok" (when the blood goes amok), which will address B-lymphocyte differentiation and haematological disorders derived from B-lymphocytes. Alex Schmitz was granted a support (legat) of 3500 DKK to participate in the Infinicyt-Workshop, 1st of April at the conference center at Stockholm airport, Sweden.

Phd. defence announced

Linda Pilgaard will defence her phd. project with the title: "Investigating the effects of hypoxic preconditioning and reduced oxygen tension on in vitro chondrogenesis in human adipose tissue-derived stem cells (ASCs)" the 24. of April, 13.00, at Bajers Vej 3B. The work has been done at the Laboratory of Stem Cell Research, Department for Health Science and Technology with Associate Professor Vladimir Zachar as advisor.

Martin Bøgsted has attended the 4th International qPCR Event, Technische Universität München, March 9-13, 2009, and presented the talk "Design and analysis of Q-RT-PCR assays for haematological malignancies using mixed effects models" by Martin Bøgsted, Charlotte Mandrup, Anders Petersen, Steen Falgreen, Hans Erik Johnsen, Anne Bukh and Karen Dybkær.

Meeting in the laboratory group of The Danish Myeloma Study Group (DMGS)

The initial goal of DMGS is to establish a national biobank containing material from myeloma patients. On March 4th, the laboratory subgroup of DMGS meet at the Department of Clinical Pathology, Odense University Hospital to discuss the practical details of collecting, handling and storing the biologic material. From the Haematology BioBank, Linda Pilgaard and Torben L. Andersson participated.

Participation in The European Myeloma Network Gene Expression Profiling workshop 30.-31. January 2009 in Rotterdam.  

Anne Dirks, Martin Bøgsted, Kim S. Bergkvist, Hans E. Johnsen and Linda Pilgaard  took part in the workshop where the aim was to discuss viewpoints on gene expression profiling by microarray from preparation to analysis from myeloma study groups across Europe, including the groups of Dirk Hose, Antonino Neri, Gareth Morgan, Herve Avet-Loiseau, Hans Johnsen and Pieter Sonneveld.

13/01/2009 Scholarship from "Kræftens Bekæmpelse"

Master student in Medical Biotechnology, Emilie Kallehauge, has received a scholarship of 70.000 DKK from "Kræftens Bekæmpelse" for the project: "Regulation of phenotypic characteristics by microRNAs in diffuse large B-cell lymphomas".

The aim of the project is to study the expression of three specific microRNAs which is differentially expressed in normal lymphoid tissue, DLBCL cell lines, and DLBCL tumours by qPCR and global microRNA arrays. The effect of the microRNAs overexpression or down regulation on cancer relevant processes as apoptosis, cell cycle and proliferation will be examined by transfection mediated knockdown or overexpression studies of each microRNA in DLBCL cell lines.
